Digital transformation is rapidly reshaping pharmaceutical research, formulation development and manufacturing through the integration of advanced digital technologies under the emerging paradigm of Digital Pharmaceutics and Pharmaceutical Industry 4.0. Traditional pharmaceutical development relies extensively on experimental approaches, which may require substantial time, resources and iterative optimisation. Digital technologies, including artificial intelligence, machine learning, Internet of Things, big data analytics, cloud computing, digital twins, robotics and process automation, offer opportunities to improve formulation design, process understanding, quality control and manufacturing efficiency. Artificial intelligence and machine learning can facilitate prediction of drug–excipient interactions, formulation performance, dissolution behaviour and critical quality attributes, thereby supporting data-driven formulation optimisation. The integration of Quality by Design and Process Analytical Technology with digital systems enables real-time monitoring, predictive process control and improved product consistency. Digital twins and connected manufacturing platforms further support simulation, process optimisation, predictive maintenance and continuous manufacturing. Despite these advantages, implementation remains associated with challenges involving data integrity, cybersecurity, interoperability, regulatory validation, infrastructure investment and workforce competencies. This review critically examines the role of digital technologies in modern pharmaceutical formulation and manufacturing, with particular emphasis on artificial intelligence, machine learning, IoT, big data, digital twins, QbD, PAT and continuous manufacturing. The review also discusses current applications, benefits, limitations, regulatory considerations and future prospects for developing intelligent, connected and patient-centric pharmaceutical manufacturing systems.
